Seed transfer operation
The seed transfer pipeline should be sterilized before the seed was shifted from seed tank to fermentation tank (fer-tank). The operation step is as following.
Two harvest valves are connected firmly by a soft pipe with a bypass with a valve which can endure the pressure of 0.4MPa. A bypass with a valve was mounted on the pipe.
Open the V08 and V09 both tank in turns, then the condensate would drained from bypass, when the pressure of pipe is 0.1~0.2 , time was recorded.
After the pipe was sterilized for about 30~60 minutes, decrease the pressure of fer-tank to 0.02MPa,
a. If the transfer pipeline was employed for inoculation, then open the inoculate diaphragm valve, close the steam valve of fer-tank, and close the bypass value, and seed tank V08 valve. After the steam in the inoculating pipe entered fer-tank, then promoted the fer-tank pressure at 0.06~0.08MPa, and opened the bottom valve of seed tank, then decreased the pressure of fer-tank at 0.02MPa to let the seed into fer-tank. When the seed amount was enough, closed the bottom valve of fer-tank.
b. If the harvest valve was employed for inoculation, then open the V09, close V08 of fer-tank, and close the bypass value, and the V08 valve of seed tank. After the steam in the inoculating pipe entered fer-tank, then promoteed the fer-tank pressure at 0.06~0.08MPa, and opened the bottom valve of seed tank, then decreased the pressure of fer-tank at 0.02MPa to let the seed into fer-tank. When the seed amount was enough, closed the bottom valve of fer-tank
Adjust the pressure and air flux to meet the request, and then setup other parameter according with “controlling system operation instructor”. The fermentation process began after inoculated, the parameters, such as temperature, pH, DO, weight etc, would be recorded by computer automatically.
Closed fer-tank harvest valve(V09), then took inoculating pipe away, then sterilized the sampling pipeline with steam for 20~30 minutes, then close the valve of V09 and V08. |
